Hunter, Kelly M.;Wagner, Jackson C.;Kalaj, Mark;Cohen, Seth M.;Xiong, Wei;Paesani, Francesco research published 《 Simulation Meets Experiment: Unraveling the Properties of Water in Metal-Organic Frameworks through Vibrational Spectroscopy》, the research content is summarized as follows. In nanoporous materials, host-guest interactions affect the properties and function of both adsorbent and adsorbate mols. Because of their structural and chem. diversity, metal-organic frameworks (MOFs), a common class of nanoporous materials, have been shown to be able to efficiently and, often, selectively adsorb various types of guest mols. In this study, we characterize the structure and dynamics of water confined in ZIF-90. Through the integration of exptl. and computational IR (IR) spectroscopy, we probe the structure of heavy water (D2O) adsorbed in the pores, disentangling the fundamental framework-water and water-water interactions. The exptl. IR spectrum of D2O in ZIF-90 displays a blue-shifted OD-stretch band compared to that of liquid D2O. The anal. of the IR spectra simulated at both classical and quantum levels indicates that the D2O mols. preferentially interact with the carbonyl groups of the framework and highlights the importance of including nuclear quantum effects and taking into account Fermi resonances for a correct interpretation of the OD-stretch band in terms of the underlying hydrogen-bonding motifs. Through a systematic comparison with the exptl. spectra, we demonstrate that computational spectroscopy can be used to gain quant., mol.-level insights into framework-water interactions that determine the water adsorption capacity of MOFs as well as the spatial arrangements of the water mols. inside the MOF pores which, in turn, are key to the design of MOF-based materials for water harvesting.

Huang, Lingyun;Yang, Yinxin;Wu, Ruiyao;Fan, Weifeng;Dai, Quanquan; He, Jianyun;Bai, Chenxi research published 《 Boron nitride and hyperbranched polyamide assembled recyclable polyisoprene vitrimer with robust mechanical properties, high thermal conductivity and remoldability》, the research content is summarized as follows. Rubber has huge potential applications for thermal management due to its outstanding softness and elasticity, however, its low thermal conductivity limits its applications, and the vulcanized rubber also has the problem of being unrecoverable. Therefore, the development of recyclable rubber-based material with good mech. properties, high thermal conductivity remains a challenge. Herein, the dynamic β-hydroxyl ester bonds were introduced into the composites of the epoxidized boron nitride (EBN) and the epoxidized polyisoprene (EPI) matrix via carboxyl-terminated hyperbranched polyamide (HBPA), forming dynamic covalently crosslinking rubber-based vitrimers. The covalent-bonds-mediated interface reduces the phonon scattering and enables the material to have a relatively high thermal conductivity of 0.51 W/mK under a loading of 50 phr (parts per one hundred parts of rubber) of EBN. In addition, the introduction of exchangeable bonds brings the materials the capability to change the network topol. at high temperatures, exhibiting excellent recyclability and remoldability.

Huang, Lingyun;Yang, Yinxin;Niu, Zhen;Wu, Ruiyao;Fan, Weifeng;Dai, Quanquan; He, Jianyun;Bai, Chenxi research published 《 Catalyst-Free Vitrimer Cross-Linked by Biomass-Derived Compounds with Mechanical Robustness, Reprocessability, and Multishape Memory Effects》, the research content is summarized as follows. Vitrimerization of thermoset polymers plays an important role in addressing resource recovery and reuse. Vitrimer elastomers with good mech. properties often require well-designed crosslinking agents or fillers, but this increases processing complexity or reduces vitrimer dynamic properties. In this report, a simple green strategy to build a strong vitrimer elastomer is designed. Com. available epoxidized natural rubber (ENR) is crosslinked with biomass-derived D-Fructose 1,6-bisphosphoric acid to get a vitrimer elastomer crosslinked by β-hydroxy phosphate ester bonds and has abundant hydrogen bonds. Hydrogen bonds can preferentially break and dissipate energy under external forces, which makes the sample robust. The topol. network can be reformed at high temperatures through the dynamic exchange of β-hydroxy phosphate ester bonds, which gives the material malleability and recyclability. In addition, through the strategy of combining reprocessing and welding, multiple shape memory effects can be achieved in one postprocessing step. Considering that a variety of com. available epoxy polymers are easily available, it is believed that this strategy can be a simple and versatile way to enable com. epoxy polymers to achieve green crosslinking through biomass crosslink agents, which results in robust and recyclable vitrimers based on β-hydroxy phosphate bonds.

Hu, Xiao;Zhang, Runan;Wemyss, Alan M.;Elbanna, Mohamed A.;Heeley, Ellen L.;Arafa, Mustafa;Bowen, Chris;Wang, Shifeng;Geng, Xueyu;Wan, Chaoying research published 《 Tuning triboelectric and energy harvesting properties of dielectric elastomers via dynamic ionic crosslinks》, the research content is summarized as follows. The bromination of poly(isobutylene-co-isoprene) rubber introduces a small amount of bromide groups (1-2 mol%) to the elastomer backbone and creates new opportunities for functionalisation, as compared to other saturated and diene elastomers. In this work, three types of nucleophile reagents: namely pyridine, triphenylphosphine and imidazoles bearing four types of side groups of Me, Et, hydroxyl or vinyl group were introduced to brominated poly(isobutylene-co-isoprene) rubber (BIIR) through nucleophile substitution with the bromine via solid-state rubber compounding and curing processes. The resulted ionic aggregates act as phys. crosslinks and their size and d. directly affected the mech. reinforcement, self-healing and dynamic mech. properties of the elastomers. The smaller and polar imidazolyl/bromine pairs led to the highest reinforcement beyond even the sulfur-cured BIIR counterparts. The 1-Et imidazole (EIm) modified BIIR showed the highest tensile strength of 17.01 ± 1.89 MPa and elongation at break of 1402 ± 69% with self-healing efficiency of 63.7%, after being treated at 140°C for 30 min. In addition, the inclusion of the ionic clusters enhanced the relative permittivity of the elastomer, thereby enhancing the energy conversion efficiencies. The nucleophile substitution reaction via conventional solid-state rubber compounding processes provides a facile crosslinking and reinforcement strategy for halogen-containing polymers. In addition, the dynamic ionic crosslinking networks spontaneously benefit electromech. and self-healing properties of the dielec. elastomers.

Hu, Ming-Hao;Yu, Bing-Ying;Wang, Xiaodong;Jin, Guangyi research published 《 Drug-like biimidazole derivatives dually target c-MYC/BCL-2 G-quadruplexes and inhibit acute myeloid leukemia》, the research content is summarized as follows. Chemotherapy is the main approach for treating acute myeloid leukemia (AML). However, this therapy can cause severe side effects as well as drug resistance, hence calling for new therapeutic strategies. As c-MYC and BCL-2 are often overexpressed in AML, and synergism between c-MYC and BCL-2 promotes tumorigenesis, therefore, dual targeting of c-MYC/BCL-2 promoter G-quadruplexes (G4s) and then inhibiting the targeted gene expression would be a potential strategy in ALM treatment. In this work, in the search of dual ligands, we performed a screening assay with an inhouse, imidazole-based compound library. Consequently, two drug-like biimidazole derivatives were identified as selective c-MYC/BCL-2 G4 binders, of which, BIM-2 was selected as the candidate for inhibiting AML cell growth. Then, BIM-2 was demonstrated to downregulate both c-MYC and BCL-2 expression, and thereby cause cell cycle arrest at G0/G1 phase and apoptosis in AML cells. Furthermore, the possible end-stacking binding modes between BIM-2 and c-MYC/BCL-2 G4s were revealed by NMR and mol. docking studies. Accordingly, this study provides a new class of drug-like dual-selective c-MYC/BCL-2 G4 ligands for the potential treatment of AML.

A class of rotaxane is created, not by encapsulating a conventional linear thread, but rather by wrapping a large cucurbit[10]uril macrocycle about a three-dimensional, cylindrical, nanosized, self-assembled supramol. helicate as the axle. The resulting pseudo-rotaxane is readily converted into a proper interlocked rotaxane by adding branch points to the helicate strands that form the surface of the cylinder (like branches and roots on a tree trunk). The supramol. cylinder that forms the axle is itself a member of a unique and remarkable class of helicate metallo-drugs that bind Y-shaped DNA junction structures and induce cell death. While pseudo-rotaxanation does not modify the DNA-binding properties, proper, mech.-interlocked rotaxanation transforms the DNA-binding and biol. activity of the cylinder. The ability of the cylinder to de-thread from the rotaxane (and thus to bind DNA junction structures) is controlled by the extent of branching: fully-branched cylinders are locked inside the cucurbit[10]uril macrocycle, while cylinders with incomplete branch points can de-thread from the rotaxane in response to competitor guests. The number of branch points can thus afford kinetic control over the drug de-threading and release.

Methimazole inhibits activation of the IFN-g-induced Janus kinase (JAK)/STAT signaling pathway in FRTL-5 thyroid cells, which may account for its immunodolulatory effects. Additionally, methimazole is an inhibitor of thyroperoxidase.

Methimazole is a thiourea antithyroid agent that prevents iodine organification, thus inhibiting the synthesis of thyroxine. Antihyperthyroid.

Methimazole is an inhibitor of thyroid hormone synthesis. It is a substrate for thyroid peroxidase that traps oxidized iodide, preventing its use by thyroglobulin for thyroid hormone synthesis. Methimazole (0.4 mg/kg) inhibits the absorption of radiolabeled iodide by the thyroid gland in rats by 80.9%.3 It reduces the incidence of lymphocytic thyroiditis in the insulin-dependent type 1 diabetic BB/W rat. Methimazole has been used to induce hypothyroidism in mice. Formulations containing methimazole have been used in the treatment of hyperthyroidism.

Methimazole is a thyreostatic compound, and an antihormone, which is widely used in medicine for the treatment of hyperthyroidism.

Methimazole is a thioamide inhibitor of the enzyme thyroid peroxidase (TPO), with antithyroid activity. Upon administration, methimazole inhibits the metabolism of iodide and the iodination of tyrosine residues in the thyroid hormone precursor thyroglobulin by TPO; this prevents the synthesis of the thyroid hormones triiodothyronine (T3) and thyroxine (T4).

Methimazole is an antithyroid medication which is now considered the first line agent for medical therapy of hyperthyroidism and Graves disease. Methimazole has been linked to serum aminotransferase elevations during therapy as well as to a clinically apparent, idiosyncratic liver injury that is typically cholestatic and self-limited in course.
Methimazole, also known as tapazole or danantizol, belongs to the class of organic compounds known as imidazolethiones. Hiremath, Chaitanya N. research published 《 Abbreviated Profile of Drugs (APOD): modeling drug safety profiles to prioritize investigational COVID-19 treatments》, the research content is summarized as follows. Safe and effective oral formulation of a drug that is easy to store, transport, and administer, is imperative to reach the masses including those without adequate facilities and resources, in order to combat globally transmitted coronavirus disease 2019 (COVID-19). In this decision analytic modeling study, the safety of investigational COVID-19 drugs in clin. trials was assessed using the Abbreviated Profile of Drugs (APOD) methodol. The method was extensively tested for various unbiased datasets based on different criteria such as drugs recalled worldwide for failing to meet safety standards, organ-specific toxicities, cytochrome P 450 inhibitors, and Food and Drug Administration (FDA) approved drugs with remarkable successes. Exptl. validation of the predictions made by APOD were demonstrated by comparison with a progression of multiparametric optimization of a series of cancer drugs that led to a potent drug (GDC-0941) which went into the clin. development. The drugs were classified into three categories of safety profiles: strong, moderate and weak. A total of 3556 drugs available in public databases were examined According to the results, drugs with strong safety profiles included molnupiravir (EIDD-2801), moderate safety profiles included dexamethasone, and weak safety profiles included lopinavir. In this anal., the physicochem.-pharmacokinetic APOD fingerprint was associated with the drug safety profile of withdrawn, approved, as well as drugs in clin. trials and the APOD method facilitated decision-making and prioritization of the investigational treatments.
